Cytotoxic Effects of Actinomycin D, Adriamycin and Puromycin on the Development Stage of Early Mouse Embryos

Abstract
The present experiments were undertaken in order to elucidate the cytotoxicity of several cancer chemotherapeutic agents (Actinomycin D, Adriamycin and Puromycin) on the development of mouse early embryos in vitro.
Two-cell embryos were cultured for 72 hours in the absence or presence of various concentrations of Actinomycin D, Adriamycin and Pu:-omycin by adopting two culture system, the micro tube culture method and paraffin drop culture method and biochemical observation was also performed to examine the changes of membrane transport and protein synthesis of ambryos.
The 2-cell stage exposed to Actinomycin D, Adriarnycin and Furomycin for 24 hours respectively was labeled with ¢¥H-lencine for 2 hours. The following conclusions were obtained :
1. The 2-cell stage was remarkably inhibited by concentration of Actin - ornycin D of D. 1pg/ml in cleavage of embryonal cells when 2-cell embryos were cultured for 72 hours in various concentrations of Actinomycin D.
2. Adriamycin suppressed severely the development of embryos at concentration of 0.2pg/ml.
3. Puromycin inhibited markedly cleavage of embryonal cells at concentration of 45pg/mI.
4. Actinomycin D and Puromycin suppressed both the uptake and incorporation of precusors for protein synthesis in damaged embryonal cells but Adriamycin didn¢¥t give an influence on the uptake of precusor but suppressed the incorporation of precusor, ¢¥H-leucine.
From the above results, it seems to indicate that Actinomycin D, Adriamycin and Puromycin inhibit the development of mouse embryo by suppressing the protein synthesis which is essential for cleavage of embryonal cells.
